How I Learned to Streak Bacteria to Isolate Single Colonies

First, we must understand what makes a student proficient in this area. A student is proficient if they: demonstrate the ability to streak bacteria to isolate a single colony, uses aseptic technique, the plate has no contamination and has distinct colonies, the streaking pattern is correct, and they can aseptically inoculate a plate with a liquid culture.


Now that we know what makes a student proficient, we can begin to talk about where I have applied these skills. In the pGLO transformation lab, I had to streak plates and isolate colonies. In order to streak plates aseptically, you must hold the plate lid while hovering the lid over the plate as you rotate the plate
